Synthesis of (±)－2－(p－Haloaryl)－2－hydroxyacetic acid Derivatives

Abstract

|
2-Hydroxyacetic acids are often used as starting compounds in organic synthesis. There are a lot of synthetic methods of 2-hydroxyacetic acid derivatives. but often it is not easy to synthesize some derivatives.
In this study condensation reaction of diethyl oxomalonate with aromatic hydrocarbons was applied. In the condensation reaction of 4′-halo-1.1′-biphenyls (Cl. Br) with diethyl oxomalonate in the presence of tin(Ⅳ) chloride diethyl 2-〔(4′-halo-1.1′-biphenyl)-4-yl〕-2-hydroxymalonates were formed and then the corresponding esters were hydrolyzed with potassium hydroxide to the 2-hydroxydicarboxylic acids. By decarboxylation of the corresponding 2-hydroxymalonic acids in c-HCl at 100℃ the final products (±)-2-〔(4′-halo-1.1′-biphenyl)-4-yl〕-2-hydroxyacetic acids were prepared.
